1. Objective
To demonstrate, with reproducible data, that the defined shipping system holds the product within its labeled storage range <<FILL: e.g. 2 to 8 degrees C>> for at least the target hold duration <<FILL: e.g. 96 hours>> when exposed to defined worst-case seasonal ambient temperature profiles, at both minimum and maximum payload, using coolant conditioned to a controlled specification.
2. Scope
This protocol qualifies one frozen shipper configuration: a fixed bill of materials (insulation, coolant type and mass, pack-out arrangement) tested against the seasonal profiles below. It does not qualify a transport lane; lane performance is confirmed separately (see the lane qualification protocol). Any change to insulation, coolant type or mass, conditioning specification, or pack-out arrangement invalidates this qualification and requires requalification under <<FILL: SOP-ID for change control>>.

4. Prerequisites
- Shipper bill of materials is frozen and under document control.
- Data loggers are calibrated, traceable to a national standard, with pre-use calibration current and post-use verification planned (accuracy

- The pack-out instruction to be tested is drafted and available (see the pack-out work instruction).
- Coolant conditioning equipment is qualified and mapped.

6. Ambient profiles under test
Qualify against a hot (summer) profile and a cold (winter) profile so both failure directions are covered: heating above the upper limit and coolant-driven freezing below the lower limit. Industry seasonal profiles such as ISTA 7D may be referenced; describe the profile you use by its ramp points, do not paste copyrighted profile text.

7. Acceptance criteria
A run passes when all of the following are true:
- Every product-side logger stays within the product range
<<FILL: e.g. 2.0 to 8.0 degrees C>>for the entire target hold duration. - No product-side logger crosses the freeze threshold for a freeze-sensitive product, even under the winter profile.
- The result is reproducible: all required replicates for a profile and payload pass, with results clustered rather than a single marginal pass.
- Coolant conditioning was verified to specification before pack-out.
- Post-use logger verification confirms drift within tolerance; otherwise the run data is invalidated.
The configuration is qualified only when every profile and payload combination meets these criteria across all replicates.

9. Deviation handling
Any failing run, invalid logger, or departure from the protocol is recorded as a protocol deviation with an assessment of impact on the conclusion. A failing run cannot be discarded as an outlier without a documented, investigated technical cause. Route deviations per <<FILL: SOP-ID for deviations>> and have QA disposition before the report is finalized.
10. Summary and conclusion
State whether the configuration met all acceptance criteria across every profile, payload, and replicate. Record the qualified claim (range, hold duration, payload window, profiles qualified), the conditions that would trigger requalification, and the controlled documents produced (validated pack-out instruction, conditioning specification). QA signs the conclusion.
11. Attachments
- Calibration certificates and post-use verification for every logger.
- Chamber run records and reference-sensor traces.
- Photo-documented pack-out for each configuration.
- Raw logger data files (retain the raw file, not only an auto-generated report).

Conclusion in the specimen: the configuration held 2.0 to 8.0 degrees C for the full 96-hour target under both seasonal profiles at minimum and maximum payload, with no freeze event, across all replicates. The pack-out instruction and the conditioning specification are locked as controlled documents; any change to insulation, coolant mass, conditioning, or pack-out triggers requalification.
Common inspection findings this protocol prevents
- Only the summer profile qualified, so a winter freeze failure mode is never tested for a freeze-sensitive biologic.
- A single passing run is presented as qualification, with no replicates and no reproducibility.
- Coolant conditioning is not verified before pack-out, so the field result does not match the qualification.
- The thermal surrogate does not represent the real product mass, so the qualification does not bound the true payload.
- Only maximum payload is tested; a near-empty box, which behaves differently, is never challenged.
- Auto-generated report retained but the raw logger file discarded, so the original record is gone.
How to adapt this protocol
- Set your document number, product family, storage range, and target hold duration on the approval page.
- Enter the real bill of materials and both payload configurations in section 3.
- Define your seasonal profiles in section 6 by ramp points; reference an industry profile by name only, do not paste its text.
- Set the replicate count in section 8 from your risk justification (three per combination is common).
- Point the deviation and change-control cross-references to your real procedures.
- Confirm the current version of every referenced standard before issue.
