Every product that goes through a GxP process depends on materials, services, and systems provided by external organizations. Raw materials, APIs, excipients, packaging components, contract manufacturing, contract testing, software systems, calibration services, all of them are supplied by vendors who are not directly under your quality management system but whose quality directly affects your product quality.
Supplier qualification is the process of establishing that a supplier is capable of consistently providing materials or services that meet your requirements and applicable GxP standards. It’s a regulatory requirement, not an optional best practice. 21 CFR 211.84 requires testing and approval of drug components from suppliers. 21 CFR 211.68 requires computer system validation, and for software vendors, that includes qualification of the supplier.
The Approved Vendor List (AVL)
The AVL (also called the Approved Supplier List or ASL) is the controlled list of suppliers that have been qualified to supply materials or services to your GxP operations. Only approved vendors on the AVL can supply GxP materials or services.
The AVL is not just a list. It’s a controlled document that includes:
- Supplier name and contact information
- Materials or services supplied
- Qualification status (approved, conditional, suspended, disqualified)
- Qualification date and method
- Required qualification level
- Next scheduled audit or review date
Adding a supplier to the AVL requires a formal qualification process. Receiving materials from an unapproved supplier is a GMP violation. Using an unapproved supplier for contract testing is a GCP violation if it affects clinical data. These aren’t technicalities.
Qualification Levels
Not every supplier needs the same level of qualification. Risk-based tiering is the standard approach:
Tier 1: Critical Suppliers Suppliers providing materials or services that directly and significantly affect product quality, patient safety, or data integrity. Examples: API manufacturers, contract manufacturing organizations (CMOs), contract testing laboratories (CTLs), critical excipient suppliers, validated system software vendors.
Qualification requirements: Full supplier questionnaire, complete quality system documentation review, on-site audit before approval, Quality Agreement, ongoing monitoring including periodic re-audit.
Tier 2: Major Suppliers Suppliers providing materials or services with significant GxP impact but less direct patient safety risk. Examples: excipient suppliers, packaging component suppliers, calibration service providers, EDMS vendors.
Qualification requirements: Questionnaire, documentation review, possibly desk audit or scheduled on-site audit. Quality Agreement.
Tier 3: General Suppliers Suppliers of indirect materials (laboratory consumables, office supplies) or services with minimal GxP impact.
Qualification requirements: Basic questionnaire, certificate of compliance review, no audit typically required.
The Qualification Process
Step 1: Supplier Identification and Initial Screening Identify the need and candidate suppliers. Collect basic information: company profile, certifications (ISO 9001, ISO 13485, GMP license, FSMA registration), regulatory inspection history, any warning letters or consent decrees.
For high-risk suppliers, check FDA’s warning letter database and Establishment Inspection Report (EIR) availability. A supplier with a recent critical warning letter related to the materials you’re buying is a significant risk.
Step 2: Supplier Questionnaire A structured questionnaire collects detailed information about the supplier’s quality system: organizational structure, quality management system scope and certification, batch release process, documentation and record retention practices, change control procedures, deviation and CAPA management, environmental controls, contamination prevention, and regulatory inspection history.
Design the questionnaire to get the information you actually need, not a generic document that every supplier fills out the same way. Questions about the specific materials or services you’re procuring are more valuable than generic quality system questions.
Step 3: Documentation Review For Tier 1 and 2 suppliers: review quality system documentation. This includes: quality manual, QMS certificates (ISO, GMP), regulatory inspection reports or certificates of GMP compliance, Certificate of Analysis formats, analytical testing records for recent batches, and the supplier’s own customer audit results if available.
For software vendors: review the Software Development Life Cycle documentation, bug/defect tracking process, release management procedures, validation documentation package (IQ/OQ template, DQ documentation), and regulatory compliance statements.
Step 4: On-Site Audit For Tier 1 suppliers, an on-site audit is required before approval. The audit evaluates the quality system in practice, not just on paper.
Audit focus areas for a contract testing laboratory:
- Analyst training records and qualifications
- Instrument calibration and qualification status
- SOP coverage and document control
- Data integrity controls (audit trails, access controls, raw data retention)
- OOS investigation process
- Environmental conditions in the laboratory
- Reference standard management
Audit focus areas for an API manufacturer:
- Manufacturing process controls
- Batch release testing and documentation
- Change control (particularly for changes that might affect impurity profile)
- Stability program
- Environmental monitoring
- Contamination controls and cross-contamination prevention
Step 5: Quality Agreement A Quality Agreement is a formal contract that defines the quality responsibilities of each party. It is required for contract manufacturers, contract testing laboratories, and certain software and service providers.
For a contract manufacturer, the Quality Agreement specifies: who is responsible for batch release, who approves the batch manufacturing record, what changes require notification, who manages regulatory submissions for the contracted activity, how deviations are reported and managed, and who has the right to inspect the facility.
For a software-as-a-service vendor (cloud LIMS, cloud EDMS), the Quality Agreement addresses: how the vendor manages updates and notifies customers, who has responsibility for data backup and recovery, what audit trail protections are in place, and how security incidents are communicated.
Step 6: Approval and AVL Update QA reviews all qualification documentation and approves the supplier for addition to the AVL. The approval is documented with a date and the scope of approval (specific materials, specific services, specific facility locations).
Ongoing Monitoring
Qualification is not a one-time event. Approved suppliers require ongoing monitoring:
Incoming material testing: Raw materials and components from approved suppliers are tested at receipt against specifications. This serves two purposes: confirming the material meets specification and detecting any degradation in supplier performance.
CoA review: For every incoming lot, review the Certificate of Analysis from the supplier. Compare their results to your specification. Look for trends in results near specification limits. A supplier whose impurity results are consistently just under the limit is a monitoring concern even if every lot passes.
Supplier change notification: Quality Agreements must require suppliers to notify you of significant changes before they occur: changes to manufacturing process, testing methods, facilities, key personnel, or any change affecting GxP quality. Changes that affect the material or service you’re buying may require re-qualification of the supplier or re-qualification of the incoming material.
Periodic re-audit: Tier 1 suppliers should be re-audited on a defined schedule (typically every 1-3 years for high-risk suppliers). Re-audit timing can be risk-adjusted: a supplier with consistently excellent performance and clean regulatory inspections can be re-audited less frequently than one with recent deviations or a changing quality system.
Regulatory intelligence: Monitor whether your suppliers are receiving FDA warning letters, recall actions, or import alerts. A warning letter for a contract testing lab related to data integrity is a quality event for your organization even if your materials weren’t the ones tested.
Disqualification and Suspension
When a supplier’s performance degrades, or a significant quality event occurs, the response should be proportionate to the risk:
Conditional approval: The supplier remains approved but under enhanced monitoring. Additional incoming testing, more frequent communication with the supplier’s quality team, or a corrective action request. Used when issues are manageable and the supplier is cooperative.
Suspension: No new orders while the issue is investigated. Used when a significant quality event is identified and the impact isn’t yet clear. Suspension prevents further risk while the situation is assessed.
Disqualification: The supplier is removed from the AVL. All inventory from the disqualified supplier is quarantined and assessed. Used when qualification failures are fundamental and cannot be remediated with confidence.
Disqualification decisions require QA approval, documentation, and a plan for managing affected inventory and transitioning to alternative suppliers.
Software Vendor Qualification
Software vendor qualification is a specialized form of supplier qualification that’s directly connected to computer system validation. When you validate a LIMS or EDMS, you’re validating your implementation of the vendor’s platform. But the platform itself rests on the vendor’s quality system.
Key elements of software vendor qualification:
- Does the vendor have a formal SDLC process?
- How are defects identified and tracked?
- How are releases tested and documented before release?
- What regulatory support documentation does the vendor provide?
- How are security vulnerabilities identified and patched?
- What is the vendor’s data retention and business continuity plan?
The output of software vendor qualification affects how much site-level validation is needed. A vendor with a mature quality system and strong regulatory documentation can justify reduced site-level testing for non-critical functions. A vendor with a limited quality system increases the site’s validation burden because the vendor’s own testing can’t be use.
For cloud and SaaS vendors, additionally assess: where is data stored (country/jurisdiction), what are the contractual protections for your GxP data, what happens to your data if the vendor goes out of business, and what audit rights do you have as a customer?
Supplier Qualification for Critical Raw Materials in Biologics and CGT
In biologics and CGT manufacturing, raw material supplier qualification takes on additional complexity because:
- Biological raw materials (cell culture media, bioreactor consumables, viral vector production reagents) can have batch-to-batch variability that directly affects process performance
- GMP-grade raw materials for CGT (growth factors, serum alternatives, plasmids, viral vectors) have limited supplier options and may involve long qualification timelines
- Supply chain disruption risk is higher for specialized materials with few qualified suppliers
Raw material qualification for CGT may include: qualification of the supplier’s manufacturing facility, testing of incoming lots for process-relevant performance (not just chemical specification), and bridging studies when a new supplier lot is introduced.
The data integrity requirements for raw material testing in CGT are the same as for any other GxP testing, but the consequence of a raw material failure is potentially catastrophic: for an autologous CGT product, the patient’s cells may be the only starting material available. Getting it right the first time is essential.
